NR vs NRH

Just like NMNH, NHR is the reduced form of NR. Classic NR is typically bonded to chloride, and to figure out if NRH is a more stable molecule than NR, researchers did two experiments. In the first, they injected NRH and in the second they give NRH orally in mice. In the first experiment, after two hours, NR was degraded, while NHR was far more stable. When given orally, NRH was detected after 1 hour in the plasma, unlike NR levels that were undetectable.

During studies, NRH was found to be a far more stable and better molecule, compared to NR, which is degraded to nicotinamide. When orally administrated, NHR is a more potent and faster NAD+ booster than NR in mammalian cells and tissues.

NMNH vs NMN

NAD+ is an essential molecule for optimal cellular functions. As we age, NAD+ levels decrease. The body gets NAD+ replenishment by supplementation with NAD+ precursors. NMNH increases NAD+ levels almost 10 times at lower concentrations than that needed for NMN. Also, NMNH has reached a plateau at high concentrations and rises NAD+ levels 10-fold with supplementation, while NMN was able to double the NAD+ levels in these cells and reached a plateau after only 1 hour. NMNH has significantly increased NAD+ levels within 15 minutes and stayed stable for 24 hours.

Administration of NMNH can also increase other NAD+– related metabolites, such as nicotinic acid adenine dinucleotide (NAAD), which is found to be a marker for increasing NAD+ levels.

In vivo, administration of NMNH can increase NAD+ levels far more efficiently in the blood than in NMN. Supplementation with NMN appeared to decline NAD+ levels in the blood after 4 hours of the first injection, while NMNH double the NAD+ levels and was stable up to 20 hours.

According to all these studies and results, NMNH is the new potent form of increasing NAD+ far more efficient and faster than other NAD+ precursors, both in vitro and in vivo. While NMN has failed, NMNH can be used as a potential therapeutic agent, because it can increase NAD+ levels in other tissues and be safe after a long-term administration.

NADH conversion into NMNH and AMP was performed with incubation of EcNADD (enzymes of Escherichia Coli) with NADH and Tris HCL. The separation of NMNH from AMP was reached with the HPLC method, by C18 reverse-phase chromatography. Pure NMNH fractions were evaporated until dry.
