NMNH Powder: Reduced NMN Supplement as the Most Potent NAD Booster

reduced NMN bulk powder

Reduced nicotinamide mononucleotide (NMNH) is a dietary supplement ingredient that acts as an NAD+ booster with anti-aging and longevity benefits. Taking NMNH as a supplement is known to enhance NAD+ production, suppress glycolysis, TCA cycle, and cell growth. It is also known to be effective in increasing NAD+ levels in the kidney, as well as protecting renal proximal tubular cells from hypoxia, which is an important part of acute kidney injury (AKI).

What is NMNH?

NMNH is the new and reduced form of nicotinamide mononucleotide (NMN). NMNH is newly discovered and seems to increase NAD+ more effectively than other NAD+ precursors. Studies suggest NMNH is a far more powerful NAD+ precursor.

Other terms referring to NMNH include reduced NMN, reduced β-NMN, reduced β-Nicotinamide -monophosphate, reduced Nicotinamide-1-ium-1-β-D-ribofuranoside 5′-phosphate. The CAS number for NMNH is 108347-85-9 and its molecular formula is C11H17N2O8P. NMNH appears as a white crystalline powder with a molecular weight of 335.23 g/mol.

NR vs NRH

Just like NMNH, NHR is the reduced form of NR. Classic NR is typically bonded to chloride, and to figure out if NRH is a more stable molecule than NR, researchers did two experiments. In the first, they injected NRH and in the second they give NRH orally in mice. In the first experiment, after two hours, NR was degraded, while NHR was far more stable. When given orally, NRH was detected after 1 hour in the plasma, unlike NR levels that were undetectable.

During studies, NRH was found to be a far more stable and better molecule, compared to NR, which is degraded to nicotinamide. When orally administrated, NHR is a more potent and faster NAD+ booster than NR in mammalian cells and tissues.

The molecular formula of NHR is C11H16N2O5, while the molecular formula of NR is
C11H15N2O5+.

NMNH vs NMN

NAD+ is an essential molecule for optimal cellular functions. As we age, NAD+ levels decrease. The body gets NAD+ replenishment by supplementation with NAD+ precursors. The most effective precursors were NMN and NR, but according to the studies, they both had limitations in boosting doses. High doses are needed to get the high NAD+ levels. Recent studies discover the reduced form of NMN (NMNH) as a new potent form to increase NAD+ levels more efficiently. Giving NMN and NR increase NAD+ levels in the blood, but there was no apparent effect in other tissues.

Other terms referring to NNM include Nicotinamide D-ribonucleotide, Nicotinamide ribonucleoside 5′-phosphate, Nicotinamide nucleotide, β-Nicotinamide ribose monophosphate. The CAS number of NMN is 1094-61-7, with molecular formula C11H15N2O8P.

Giving NMNH and NMN in mice in different concentrations has shown that NMNH is a better NAD+ enhancer than NMN. NMNH increases NAD+ levels almost 10 times at lower concentrations than that needed for NMN. Also, NMNH has reached a plateau at high concentrations and rises NAD+ levels 10-fold with supplementation, while NMN was able to double the NAD+ levels in these cells and reached a plateau after only 1 hour. NMNH has significantly increased NAD+ levels within 15 minutes and stayed stable for 24 hours.

Administration of NMNH can also increase other NAD+– related metabolites, such as nicotinic acid adenine dinucleotide (NAAD), which is found to be a marker for increasing NAD+ levels.

In vivo, administration of NMNH can increase NAD+ levels far more efficiently in the blood than in NMN. Supplementation with NMN appeared to decline NAD+ levels in the blood after 4 hours of the first injection, while NMNH double the NAD+ levels and was stable up to 20 hours.

According to all these studies and results, NMNH is the new potent form of increasing NAD+ far more efficient and faster than other NAD+ precursors, both in vitro and in vivo. While NMN has failed, NMNH can be used as a potential therapeutic agent, because it can increase NAD+ levels in other tissues and be safe after a long-term administration.

NMNH powder Manufacturing Process

NADH conversion into NMNH and AMP was performed with incubation of EcNADD (enzymes of Escherichia Coli) with NADH and Tris HCL. The separation of NMNH from AMP was reached with the HPLC method, by C18 reverse-phase chromatography. Pure NMNH fractions were evaporated until dry. The obtained powder was redissolved in pure water. With this last step, samples were dried to obtain NMNH as an amorphous yellow powder.

Benefits of NMNH

The following are among the benefits of using NMNH supplementation:

NAD+ support

In vitro studies showed significantly higher NAD+ levels in mice and other tissues like liver, kidney, brain, heart, and more. As new discovered NAD+ precursor, it can increase the NAD+ levels more efficiently and quickly compared to NR and NMN. The reduced NAD+ precursors can be very potent NAD+ enhancers and boosting molecules.

Anti-aging

NAD+ in our body supports DNA repair and activates our sirtuins proteins. It also facilitates many bodily processes. As we age, our NAD+ levels in our bodies decline significantly, which leads to various aging-related conditions. Because NMNH can be more efficient in raising NAD+ levels, that means that it is related to slowing down aging.

Longevity

NMNH shows to increase NAD+ levels in many cells and tissue. As an NAD+ precursor, giving as supplementation can extend both healthspan and lifespan. Previous studies imply NMNH can activate sirtuins proteins through increasing NAD+ levels. Sirtuins are responsible for longevity and can prolong lifespan in mice. Thus, NMNH has a positive impact on longevity.

NMNH powder side effects

Given NMNH as a supplement shows no side effects, and toxicity even at high doses. Therefore, it is safe to use.

NMNH powder dosage

Taking as a dietary supplement, the recommended intake is between 50-1000mg.
